Phenom II X2 555BE - When to unlock......

I have a new build with an ASUS motherboard that has CPU core-unlocking abilities, and an AMD phenom II X2 555BE processor. I have not found an answer concerning when to attempt the unlock of the two dormant cores....... Do I toggle the onboard unlock switch before I power up for the first time, or do I do I get it up and running with components and OS running smoothly before I unlock?

Just wondering.........
